Nitric oxide synthase inhibition with L-NAME ameliorates nicotine- induced reproductive organ decrease in male rat

چکیده
Testicular integrity is compromised during illness or infection leading to temporary or permanent infertility and studies have associated elevated nitric oxide (NO) with infertility. This study was designed to investigate the effect of inhibiting nitric oxide synthase on nicotine-induced reproductive organ decrease in male rats. Forty-eight male Wistar rats (160-180g) were randomly assigned to six groups and treated orally for 30 days with saline (control), nicotine (0.5mg/kg, 1.0mg/kg) with or without N Nitro-L-Arginine Methyl Ester (LNAME, 50mg/Kg). At the end of the experiment, the animals were sacrificed and their productive organs were removed and weighed immediately. There was no significant difference in the mean body weight of the experimental group. Testicular and epididymal weight was significantly decreased in the nicotine treated groups. Serum testosterone level was also significantly decreased in a dose-related manner in nicotine-treated groups. However, co-treatment with L-NAME effectively reversed the nicotine-mediated alterations in weight of reproductive organs and testosterone when compared to nicotine only. Taken together, the present data indicate the abilities of L-NAME to ameliorate nicotine-induced testicular and epididymal alteration in male rats.

Role of L-NAME, a nitric oxide synthase inhibitor, in the improvement of morphine-induced amnesia induced by nicotine
Introduction: Drugs of abuse such as nicotine and morphine used systemically by addicts produce their effects via the mesolimbic dopaminergic pathway. Furthermore, evidence indicates that some behavioral effects of nicotine and morphine are mediated by nitric oxide (NO). Based on these observations, the aim of the present study was to investigate the effects of intra-nucleus accumbens (NAc) ...

Background and Objective: Nitric oxide mediated some behavioral effects of nicotine and ethanol in the nucleus accumbens. In the present study, the effects of L-NAME (nitric oxide synthase inhibitor) in the nucleus accumbens on nicotine’s effect in ethanol-induced amnesia were investigated. Materials and Methods: In this experimental study 96 adult male wistar rats were used (12 groups)...
متن کاملInhibition of nitric oxide synthase activity improves focal cerebral damage induced by cerebral ischemia/reperfusion in normotensive rats
Introduction: Nitric oxide seems to play a dual role in ischemia/reperfusion injury. Few studies have investigated whether it exacerbates or improves brain edema. In the present study, we inhibited the activity of nitric oxide synthase by L-NAME and evaluated the cerebral infarct volume, tissue swelling and brain edema, alongside the measurement of blood flow of the ischemic region. Methods...
متن کاملINFLUENCE OF ENDO THELIUM REMOVAL AND LNAME ON RESPONSES OF RAT COMMON CAROTID ARTERY TO α-ADRENOCEPTOR AGONISTS
In this study we investigated the effects of endothelium removal and L-NAME on responses to α-adrenoceptor agonists. Male Wistar rats were killed by overdose with pentobarbitone sodium, after which the left and right common carotid arteries were removed. Rings of arteries 3-4 mm in length were cut from each vessel and then mounted in 10 mL isolated organ bath, bathed in Krebs maintained at ...

Introduction: The current study aimed at investigating the role of Nitric Oxide (NO) in the maintenance of anxiety and depression induced by stress in male Wistar rats using intra-Basolateral Amygdala (BLA) injection of NO precursor, L-arginine, Nitric Oxide Synthase (NOS) inhibitor, and L-NAME. Methods: Two 23-gauge stainless steel cannulas were placed in the BLA, stereotaxically. Seven days...
